柴金解郁安神方及其拆方对模型大鼠抑郁样行为及胃肠道功能的调节作用
Effects of Chaijin Jieyu Anshen(柴金解郁安神)Formula and Its Disassembled Formulas on Depression-Like Behavior and Gastrointestinal Function in Rats

Abstract
Objective:To study the anti-depression and gastrointestinal function-regulating effects of Chaijin Jieyu Anshen(柴金解郁安神)Formu-la and its disassembled formulae.Methods:The targets and pathways of Chaijin Jieyu Anshen Formula were predicted by chemoinformatics.Chronic unpredictable mild stress(CUMS)was adopted to establish the rat model of depression.The Jieyu(解郁)effect of the formula was evaluated by sucrose preference,open field,and Morris water maze tests.Changes in body weight,water and food intake,fecal water content,and D-xylose were measured to evaluate the Jianpi(健脾)effect of the formula.The levels of neurotransmitters[dopamine(DA),5-hydroxytryptamine(5-HT),and acetylcholine(Ach)]and inflammatory cytokines[interleukin-1β(IL-1β),interleukin-6(IL-6),and inter-leukin-10(IL-10)]in the cerebrospinal fluid and blood were determined by ELISA.RT-qPCR was employed to detect the expression of genes related to neurotransmitter and immune pathways in the hypothalamus and small intestine.The targets of neurotransmitter pathways included orexin A,orexin receptor 1(OXR1),galanin(Gal),galanin receptor 1(GalR1),galanin receptor 2(GalR2),galanin receptor 3(GalR3),dopamine receptor D1(DRD1),dopamine receptor D5(DRD5),5-hydroxytryptamine receptor 2A(HTR2A),5-hydroxytryptamine receptor 2B(HTR2B),and cholinergic receptor and muscarinic 1(CHRM1).The immune pathway targets included Toll-like receptor 4(TLR4),nu-clear factor-kappa B(NF-κB),and myeloid differentiation factor 88(MyD88).Pathological changes in the hypothalamus and small intestine were observed by HE staining.Results:Compared with the normal control group,the model control group showed decreases in food intake,wa-ter intake,body weight,sucrose preference rate,total distance,central distance,number of rearings,number of crossings,and D-xylose content and excretion rate,increases in fecal water content,total swimming distance,and average escape latency,elevations in the levels of 5-HT,DA,Ach,IL-1 β,IL-6,and IL-10 in the serum and cerebrospinal fluid,and down-regulated mRNA levels of Orexin-A,Gal,Galr1,Galr3,Drd1,Drd5,Htr1a,Htr2b,and Chrm1(P<0.05 or P<0.01).Compared with the model control group,Chaijin Jieyu Anshen Formula increased the food intake,water intake,body weight,sucrose preference rate,total distance,number of crossings,D-xylose content and excretion rate,and lev-els of 5-HT,DA,and Ach in the serum and cerebrospinal fluid,while decreasing the fecal water content,central distance,number of rearings,total swimming distance,average escape latency,and levels of IL-1β,IL-6,and IL-10 in the serum and cerebrospinal fluid(P<0.05 or P<0.01).Furthermore,Chaijin Jieyu Anshen Formula up-regulated the mRNA levels of Orexin-A,Ox1r,Gal,Galr1,Galr2,Galr3,Drd1,Drd5,Htr1a,Htr2b,and Chrm1 in the hypothalamus and small intestine(P<0.05 or P<0.01).Conclusion:Chaijin Jieyu Anshen Formula may regulate the neurotransmitter and immune pathways to play a role of Jieyujianpi(解郁健脾)in the treatment of depression due to Ganyupixu(肝郁脾虚).关键词
