加味星蒌承气颗粒对小鼠出血性脑中风后便秘的缓解作用及其电生理机制研究
Effect and Electrophysiological Mechanism of Modified Xinglou Chengqi(星蒌承气)Granules in Alleviating Constipation in Mice with Hemorrhagic Stroke

Abstract
Objective:To explore the effects and mechanism of action of modified Xinglou Chengqi(星蒌承气)Granules(XCG)in alleviating constipation in mice with hemorrhagic stroke.Methods:Sixty SPF Balb/C mice were randomly divided into a sham group,a model group,a group receiving mosapride citrate at a dose of 2 mg/kg(MC-2 group),and groups receiving XCG at doses of 8 g/kg,16 g/kg,and 32 g/kg(XCG-8,XCG-16,and XCG-32 groups).A mouse model of intracerebral hemorrhage was induced by injection of collagenases into the cau-date nucleus.Three days before modeling,mice were administrated orally with corresponding drugs or sterile water twice a day for six days.After administration,electrical mapping was performed to record the slow waves of mouse colon tissuse.In vitro primary culture of interstitial cells of Cajal(ICC)was performed,and the patch clamp technique was used to investigate the effects of modified XCG-containing serum on the pacemaker potential of ICC.Results:Compared with that in the sham group,the time of the first melena defecation was significantly de-layed,and the fecal pellet number in six hours was significantly reduced in the model group.The frequency of slow wave potentials of the proximal colon was significantly decreased in the model group(P<0.01).Compared with that in the model group,the frequency of the colon-ic slow wave potentials was increased significantly in the XCG-8,XCG-16,and XCG-32 groups(P<0.01),with the most significant difference observed in the XCG-32 group.The patch clamp results showed that the frequency of the ICC pacemaker potential was increased significantly in XCG-8,XCG-16,and XCG-32 groups(P<0.01),and the effect was concentration-dependent.Moreover,this effect could be inhibited by a hyperpolarization-activated cyclic nucleotide-gated(HCN)channel-specific blocker.Conclusion:XCG can alleviate constipation in mice with hemorrhagic stroke,and the mechanism may be related to the modulation of the pacemaker potential of ICC and the slow wave potential of the colon.关键词
