Chaihuyujinxiang Granules Regulate Gut Microbiota-Gut-Brain Axis to Improve Depression

Abstract
Objective:Based on the"gut microbiota-gut-brain"axis theory,this study explored the therapeutic mechanism of Chaihuyujinxiang(柴胡郁金香)Granules in a mouse model of depression induced by chronic unpredictable mild stress(CUMS)combined with solitary rearing.Method:A total of 72 healthy male mice were randomly divided into six groups,with 12 mice in each group.They were divided into a normal control group,a model control group,a 0.01 g/kg fluoxetine group,and three Chaihuyujinxiang Granules groups at doses of 0.3,0.6,and 1.2 g/kg.Except for the normal control group,the other groups were subjected to CUMS combined with solitary rearing and administered the cor-responding treatments daily for five weeks.After five weeks,the sucrose preference test,tail suspension test,and forced swimming test were conducted.ELISA was used to measure the levels of 5-hydroxytryptamine(5-HT),norepinephrine(NE),and brain-derived neurotrophic fac-tor(BDNF)in the hippocampus and 5-HT in the colon.The fecal microbiota structure of the mice was analyzed using 16S rDNA high-throughput sequencing technology,followed by comprehensive characterization.LEfSe analysis was performed to identify bacterial phenotypes with gut microbiota variations.The PICRUSt software was employed to predict the functional gene composition of the sample.Pearson correla-tion coefficient analysis was conducted to explore the correlation among variables.Result:Compared with those in the normal control group,mice in the model control group exhibited significantly reduced body weight,sucrose preference rate,hippocampus levels of 5-HT,NE,and BDNF,as well as decreased Nissl body count,BDNF expression levels,and colonic 5-HT levels(P<0.01).In contrast,immobility time in both the forced swimming and tail suspension tests was significantly increased(P<0.01).Analysis of gut microbiota revealed a significant in-crease in the relative abundance of Bacteroidetes,Proteobacteria,Bacteroides,Ruminococcus,and Desulfovibrio and a decrease in Firmicutes,Lactobacillus,and Helicobacter.Compared with those in the model control group,mice treated with Chaihuyujinxiang Granules showed a sig-nificant increase in body weight,sucrose preference rate,hippocampus levels of 5-HT,NE,and BDNF,Nissl body count,BDNF expression,and colonic 5-HT levels(P<0.05 or P<0.01).Additionally,immobility time in both the forced swimming and tail suspension tests was signifi-cantly reduced(P<0.05 or P<0.01).Gut microbiota analysis revealed a significant decrease in the relative abundance of Bacteroidetes,Pro-teobacteria,Bacteroides,Ruminococcus,and Desulfovibrio,along with a marked increase in Firmicutes,Lactobacillus,and Helicobacter.The results of Pearson correlation coefficient analysis revealed significant associations between depressive behaviors in mice and monoamine neuro-transmitters(5-HT and NE),neurotrophic factor(BDNF),colonic 5-HT,and gut microbiota composition.Conclusion:Chaihuyujinxiang Granules exhibit significant antidepressant effects,which are closely associated with the modulation of the"gut microbiota-gut-brain"axis.关键词
