欧前胡素通过SHP1/PDGFRβ通路抑制肝星状细胞活化的实验研究
Imperatorin Inhibits Hepatic Stellate Cell Activation via the SHP1/PDGFRβ Pathway

Abstract
Objective:To explore the effect of imperatorin on the activation of human hepatic stellate cells(LX-2)and the possible mechanism.Methods:LX-2 cells were randomly grouped as follows:transforming growth factor-β1(TGF-β1),imperatorin(10,20,and 40 μmol/L),and silybin(5 μmol/L).After drug intervention,the activation and proliferation of LX-2 cells were measured by the CCK-8 assay.Cell migration was assessed via a scratch assay.The levels of interleukin-6(IL-6)and type I collagen(Col-I)in the cell supernatant were determined by ELISA.The protein levels of α-smooth muscle actin(α-SMA),Src homology domain 2-containing protein tyrosine phosphatase 1(SHP1),and platelet-derived growth factor receptor β(PDGFRβ),as well as the phosphorylation levels of Janus kinase 1(JAK1)and signal trans-ducer and activator of transcription 3(STAT3),were determined by Western blot.Results:Compared with those in the normal control group,the LX-2 cells in the model control group exhibited a typical fibroblast-like morphological change with a long spindle shape,increased prolifer-ation and migration(P<0.01),downregulated protein level of SHP1(P<0.01),and upregulated protein levels of α-SMA,PDGFRβ,IL-6,and Col-I,as well as the phosphorylation levels of JAK1 and STAT3(P<0.05 or P<0.01).In comparison with the model control group,in-tervention with imperatorin improved the morphology of LX-2 cells,decreased the cell proliferation and migration(P<0.05 or P<0.01),up-regulated the protein level of SHP1(P<0.05),and down-regulated the protein levels of α-SMA,PDGFRβ,IL-6,and Col-I and the phospho-rylation levels of JAK1 and STAT3(P<0.05 or P<0.01).Conclusion:Imperatorin can inhibit the activation of LX-2 cells by regulating the SHP1/PDGFRβ pathway.关键词
