摘要

Abstract

Ca2+overload therapy has attracted more and more attention in the field of oncology because of its good efficacy and little side effects.however,the regulation of calcium homeostasis in cancer cells,the lack of calcium in tumor cells and the low efficiency of calcium entry into tumor cells limit its curative effect,which leads to the unsatisfactory therapeutic effect.A nano-drug loading system CAP/NAADP@ZIF-8-FA which can release acid-sensitive and magnify calcium overload was constructed,and its particle size was about 280 nm.The drug loading of capsaicin(CAP)was 20.1±1.14%,and that of nicotinic acid dinucleotide phosphate(NAADP)was 8.0±0.48%,which had obvious characteristics of drug acid sensitive release.The intracellular Ca2+content of human hepatocellular carcinoma(HepG2)cells treated with CAP/NAADP@ZIF-8-FA was about 6 times higher than that of normal cells.When the concentration of CAP/NAADP@ZIF-8-FA nanoparticles was 320 μg/mL,the inhibition rate of HepG2 cells was more than 89%,and there was no obvious toxicity to normal cells.The anti-tumor activity of CAP/NAADP@ZIF-8-FA nano-drug loading system was significantly higher than that of capsaicin alone.
