Abstract

Objective To explore and compare the therapeutic effects of wild and cultivated Polygonatum odoratum on mice with diabetic nephropathy(DN).Methods A mouse model of DN was established by feeding a high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin(50 mg·kg-1).The modeled mice were randomly divided into the normal group,the model group,the metformin group,the wild Polygonatum odoratum group,and the cultivated Polygonatum odoratum group,with 10 mice in each group.The Polygonatum odoratum groups were continuously administered the corresponding aqueous decoction of the herb by gavage for 8 weeks.After the administration,the levels of fasting blood glucose(FBG),serum urea nitrogen(BUN),serum creatinine(Scr)and 24-hour urinary protein(24 h-UTP)in each group were measured.The levels of serum interleukin(IL)-1β and IL-18 were detected by enzyme-linked immunosorbent assay(ELISA).Pathological changes of renal tissue were observed by hematoxylin-eosin(HE)staining,Masson staining,and transmission electron microscopy.The positive expression of Caspase-3 in renal tissue was measured by immunohistochemistry,and the protein expressions of Bcl-2,Bax,and Caspase-3 in renal tissue were detected by Western blot.Liquid chromatography-mass spectrometry(LC-MS)was used to analyze the components of wild and cultivated Polygonatum odoratum.Results Wild Polygonatum odoratum,cultivated Polygonatum odoratum,and metformin all significantly reduced the levels of FBG,serum BUN,serum Scr and 24 h-UTP in urine,and serum IL1β and serum IL-18 in DN mice(P<0.01,P<0.05),and repaired damaged renal cells.Furthermore,they up-regulated the protein expression level of Bcl-2(P<0.05)and down-regulated the protein expression levels of Bax and Caspase-3(P<0.05).A total of 993 chemical components under the secondary classification of the Human Metabolome Database(HMDB)were identified by LC-MS.Compared with cultivated Polygonatum odoratum,some wild Polygonatum odoratum samples contained higher contents of differential components,including fatty acids,alkaloids,amino acids,and flavonoids.Conclusion Both wild and cultivated Polygonatum odoratum have therapeutic effects on DN in mice,and the mechanism may be related to lowering blood glucose,regulating inflammation,and inhibiting cell apoptosis.Moreover,the therapeutic effect of wild Polygonatum odoratum on DN in mice is superior to that of cultivated Polygonatum odoratum.
