金线兰及其近缘种叶绿体matK基因序列分析及亲缘关系鉴定
Analysis of chloroplast matK gene sequences and phylogenetic identification of Anoectochilus roxburghii and its related species

Abstract
[Purpose]This study analyzed the polymorphism of chloroplast matK gene sequences,aiming to provide reference for the identification of Anoectochilus roxburghii and its related species.[Method]Primers were designed according to the chloroplast matK gene of A.roxburghii in GenBank,and the matK sequences of A.roxburghii and its related species were amplified.MEGA 7.0 was employed to build the neighbor-joining phylogenetic tree.The number of haplotypes,number of nucleotide polymorphic loci,haplotype diversity,and nucleotide diversity index were calculated by DnaSP 6.0.[Result]The matK sequences were obtained from 13 samples of A.roxburghii and its related species,with the lengths ranging from 1 031 bp to 1 034 bp,encoding 343-344 amino acid residues.The neighbor-joining phylogenetic tree showed that A.formosanus(A04 and A09)was closely related to A.roxburghii(Jinmai and A23).Honggan(A68)was closely related to A.koshunensis,A09,A.formosanus(EU797513 and NC061756),A04,and A.zhejiangensis.Dayuanye(A67)was closely related to A.burmannicus.The results of genetic diversity analysis showed that a total of 77 polymorphic loci were detected in the 29 matK sequences of A.roxburghii and its related species,and 21 haplotypes were defined.The haplotype diversity was 0.970,and the nucleotide diversity index was 0.017 10.[Conclusion]The matK gene exhibits high polymorphism among different species of Anoectochilus,and this gene can be utilized for the identification of the origin of Anoec-tochilus and its related species.关键词
