芒果亚洲炭疽菌RPA-LFD检测体系的建立
Establishment of the RPA-LFD Detection System for Colletotrichum asianum in Mango

Abstract
Colletotrichum asianum is an important pathogen that can cause mangoe anthracnose,seriously threatening the healthy development of the mango industry.Based on the recombinase polymerase amplification(RPA)technology,as well as specific primers and probes targeting the the genomic sequence of Colletotrichum asianum,this study established an RPA-LFD(recombinase polymerase amplication-lateral flow dipstick)detection system for Colletotrichum asianum,optimized its reaction conditions,and detected its specificity and sensitivity.The results indicated that the designed RPA-LFD detection primers Ca-RPAP1/Ca-RPAP2 and probe Ca-RPAT1 exhibited high specificity for the target fragment,the reaction condition was amplification at 38℃for 20 minutes,which could achieve specific detection of Colletotrichum asianum in mango.The detection limit of this method is 10 pg/μL,and its sensitivity is approximately ten times that of conventional PCR.The RPA-LFD detection system for Colletotrichum asianum in mango established by this research features simple operation,strong specificity and high sensitivity,and is particularly suitable for rapid pathogen detection at the grassroots or on-site.关键词
