白头翁转录组特征与SSR标记开发
Transcriptomic characterization and SSR marker development for Pulsatilla chinensis

Abstract
[Objectives]The paper aimed to develop SSR markers based on transcriptome and analyze the genetic diversity for Pulsatilla chinensis germplasm resource from the Yanshan region.[Methods]High-throughput sequencing technique was used to construct transcriptome database of P.chinensis.MISA software was used to search for SSR loci and develop SSR markers,which were used for genetic diversity analysis of 24 P.chinensis germplasm resources from the Yanshan region.[Results]65 799 Unigenes were obtained with average length of 805.14 bp.Through BLASTx analysis,Unigenes were annotated in GO,KEGG,EggNOG,Nr,SwissProt and Pfam databases with 25 602(38.91%),11 774(17.89%),24 431(37.13%),30 389(46.18%),22 001(33.44%)and 21 880(33.25%),respectively.From the P.chinensis transcriptome database,8 657 SSR loci were detected in 10 726 Unigenes,and occurrence frequency of SSR was 13.16%.Among them,the largest number was mononucleotide accounting for 44.53%,and the dominant unit was A/T.The developed 16 polymorphism primer generated 97 bands,with polymorphism rate 91.01%.The percentage of primers with polymorphism information content(PIC)>0.5 accounted for 93.75%.The UPGMA result showed that 24 P.chinensis germplasm resources from the Yanshan region were divided into two categories.[Conclusions]The SSR loci were high in frequency and polymorphism in P.chinensis transcriptome,which could provide a basis for germplasm resource analysis and molecular identification for P.chinensis.关键词
