摘要

Abstract

[Objectives]This study aimed to investigate the efficacy of tetrahydrocurcumin(THC)in reducing back-fat thickness in finishing pigs and elucidate the underlying lipid-metabolic pathways though high-throughput sequencing.[Methods]Thirty 180-day-old Duroc×Landrace×Yorkshire cross-bred barrows were randomly assigned to three treatments with single pig per pen.The control(Con)group fed the NRC diet,while the low-THC(L-THC)and high-THC(H-THC)group fed the NRC diet supplemented with 300 mg·kg-1 THC.Pigs had ad libitum access to feed and water for 28 d,after which they were electrically stunned,exsanguinated,and plasma,carcass back-fat thickness and dorsal subcutaneous adipose tissue were collected.Adipose morphology,transcriptome sequencing and Western blot analyses were performed.[Results]Dietary supplementation of THC did not affect average daily gain.Compared with the Con group,H-THC markedly reduced back-fat thickness(P<0.05)in a dose-dependent manner.HE staining showed that H-THC significantly decreased adipocyte cross-sectional area(P<0.05).THC elevated protein content in adipose tissue(P<0.05)and down-regulated FASN protein expression(P<0.05).Transcriptomic profiling revealed that THC enhanced lipid oxidation,suppressed lipogenesis,and negatively regulated the mTOR signalling pathway.Protein-protein interaction networks indicated that MAPK3,RPS5,RPL27A and POLD1 related to lipogenesis were significantly enriched in mTOR signaling,DNA metabolism,mitochondrial gene expression and protein-RNA complex organization.[Conclusion]Dietary supplementation with 300 mg·kg-1 THC could reduce back-fat thickness in finishing pigs by decreasing adipocyte size through coordinated modulation of lipid oxidation,lipogenesis,and the mTOR signalling pathway.
