参松养心胶囊调控PERK/eIF2α/CHOP通路改善阿霉素诱导的心肌损伤
Regulation of the PERK/eIF2α/CHOP pathway by Shensongyangxin capsule for alleviating adriamycin-induced myocardial injury

Abstract
AIM To investigate the effect of Shensongyangxin capsule on adriamycin-induced myocardial injury in rats and its possible mechanism by regulating protein kinase R-like endoplasmic reticulum kinase(PERK)/eukaryotic initiation factor 2α(eIF2α)/CCAAT enhancer-binding protein homologous protein(CHOP)pathway.METHODS Forty rats were randomly divided into control group,adriamycin group,Shensongyangxin capsule group and PERK inhibition group,with 10 rats in each group.WGA staining was used to detect myocardial structural changes.Masson staining and Sirius red staining were used to detect myocardial fibrosis.Serum levels of creatine kinase(CK),aspartate aminotransferase(AST),brain natriuretic peptide(BNP)and cardiac troponin T(cTnT)were detected by ELISA kits.Western blotting was used to detect protein levels of PERK,eIF2α and CHOP in myocardial tissue.Protein levels of eIF2αand CHOP in myocardial tissue were detected by immunohistochemical staining.The level of myocardial apoptosis was detected by the Tunel kit.RESULTS Compared with the control group,the myocardial cross-sectional area,fibrosis level,serum contents of CK,AST,BNP and cTnT were increased in the adriamycin group(P<0.05),the protein expressions of PERK,eIF2α and CHOP in myocardial tissue were increased(P<0.05),and the myocardial apoptosis level was increased.Compared with the adriamycin group,the myocardial cross-sectional area,fibrosis level,serum contents of CK,AST,BNP and cTnT in the Shensongyangxin capsule group were decreased(P<0.05),and the protein expressions of PERK,eIF2α and CHOP in myocardial tissue were decreased in the Shensongyangxin capsule group(P<0.05).Compared with the adriamycin group,the protein expressions of PERK,eIF2α and CHOP in the myocardium of the inhibited PERK group were decreased(P<0.05),and the apoptosis level of the myocardium was decreased in the PERK inhibition group(P<0.05).CONCLUSION Shensongyangxin capsule could down-regulate endoplasmic reticulum stress mediated by PERK/eIF2α/CHOP pathway and improve myocardial injury induced by adriamycin.关键词
