基于生物信息技术预测苦参-蛇床子治疗银屑病的研究
Prediction of Kushen-Shechuangzi in the treatment of psoriasis based on bioinformation technology

Abstract
Objective:To predict the target and pathway of Kushen(Radix sophorae flavescentis)-Shechuangzi(Fructus cnidii)in the treatment of psoriasis based on bioinformation technology.Methods:The active ingredients and targets of Kushen-Shechuangzi were screened by TCMSP.Psoriasis related targets were collected from GeneCards and DisGeNET databases.The intersection of the active ingredients targets and psoriasis targets was taken to obtain the common targets,which were potential targets.The common targets were imported into Cytoscape 3.7.2 software to construct the"TCM-active ingredients-common targets"network,and STRING was used to construct the protein-protein interaction(PPI)network of targets.Metascape database was used for enrichment analysis,and molecular docking was finally adopted for preliminary verification.Results:A total of 62 common targets of Kushen-Shechuangzi in the treatment of psoriasis were obtained,such as JUN,IL-6,ESR1,TP53,and so on.Active ingredients included quercetin,luteolin,and formononetin,which were mainly concentrated in cancer pathway,IL-17 signaling pathway,TNF signaling pathway,and Th17 cell differentiation pathway.The molecular docking results showed that quercetin,luteolin and other active ingredients were stably bound to key target proteins such as JUN and NCOA1.Conclusion:The therapeutic effects of Kushen-Shechuangzi on psoriasis may be realized through the action of quercetin,luteolin,formononetin and other ingredients on JUN,IL-6,IL-1β,CDK4 and other targetsby the regulation of IL-17 signaling pathway,TNF signaling pathway,and Th17 cell differentiation pathway.关键词
