摘要

Abstract

Cancer is a highly complex systemic disease with unique biological characteristics,and metabolic reprogramming is the key to drive tumor progression.Within the various reprogrammed metabolic pathways,the rewiring of glucose metabolism—specifically the propensity of cancer cells to utilize aerobic glycolysis(the phenomenon where glucose is preferentially fermented to lactate even under ample oxygen conditions,known as the Warburg effect)to fuel their rapid proliferation demands—has emerged as one of the most intensely researched focal areas in tumor metabolism over the past decade.Concurrently,there is growing scientific interest in the potential of Traditional Chinese Medicine(TCM)and its bioactive components to modulate aerobic glycolysis and thereby inhibit tumor initiation and development.To comprehensively synthesize the current understanding of TCM's impact on this crucial metabolic pathway,this review critically examines recent research on TCM interventions targeting aerobic glycolysis in cancer.Studies demonstrate that both TCM bioactive compounds and compound formulations can significantly influence tumor aerobic glycolysis,primarily through multi-targeted and multi-layered regulatory mechanisms:Targeting key oncogenic transcription factors c-Myc and Hypoxia-Inducible Factor-1α(HIF-1α)to inhibit their expression and activity,as these factors are master regulators of genes encoding glycolytic enzymes and transporters;Modulating the core Phosphatidylinositol 3-Kinase(PI3K)/Protein Kinase B(Akt/PKB)/Mammalian Target of Rapamycin(mTOR)signaling pathway,a central hub for cellular growth and metabolism,by disrupting downstream signaling to attenuate the promotion of glycolytic enzyme activity and expression;Regulating Non-coding RNAs(ncRNAs)to indirectly influence the transcriptional activity,mRNA stability,or translational efficiency of genes encoding key glycolytic enzymes or proteins,thereby fine-tuning their functional activity levels.Through these concerted actions,TCM bioactive components and formulations effectively suppress the abnormally heightened aerobic glycolytic flux in tumor cells,disrupting the critical energy and biosynthetic supply chains required for unchecked tumor growth and spread,and ultimately manifesting significant anti-tumor efficacy by suppressing malignant proliferation,invasion,migration,and inducing apoptosis.
