仙茅苷调节HIF-1α/VEGF信号通路对糖尿病心肌病大鼠血管生成的影响
Effect of Curculigoside on Angiogenesis in Diabetic Cardiomyopathy Rats by Regulating HIF-1α/VEGF Signaling Pathway

Abstract
Objective:To investigate the effect of curculigoside(CUR)on angiogenesis in diabetic cardiomyopathy(DCM)rats by regulating hypoxia-inducible factor-1α(HIF-1α)/vascular endothelial growth factor(VEGF)signaling pathway.Methods:DCM rat models were established by intraperitoneal injection of streptozotocin and high-fat diet feeding.Successfully modeled rats were randomly divided into DCM group,CUR low-dose group(CUR-L),CUR medium-dose group(CUR-M),CUR high-dose group(CUR-H),and PX-478 group(HIF-1α/VEGF pathway inhibitor),with 12 rats in each group.Another 12 rats served as control group.All rats underwent routine echocardiography to assess cardiac function after intervention.Fasting blood glucose(FBG),total cholesterol(TC),and triglyceride(TG)levels were analyzed.Serum lactate dehydrogenase(LDH)and creatine kinase-MB(CK-MB)were detected by enzyme-linked immunosorbent assay(ELISA).Myocardial capillary density was measured by CD31 immunohistochemistry.Myocardial tissue pathological damage was observed by hematoxylin-eosin(HE)staining.HIF-1α and VEGF protein expressions were detected by Western blot.Results:Compared with control group,DCM group showed decreased left ventricular ejection fraction(LVEF),left ventricular fractional shortening(LVFS),myocardial capillary density,and HIF-1α/VEGF expressions(P<0.05),and increased serum FBG,TC,TG,LDH,and CK-MB levels(P<0.05).Compared with DCM group,CUR-L,CUR-M,and CUR-H groups showed increased LVEF,LVFS,myocardial capillary density,and HIF-1α/VEGF expressions,and decreased serum FBG,TC,TG,LDH,and CK-MB levels(P<0.05).Compared with CUR-H group,PX-478 group showed significant reversal of these indicators(P<0.05).Conclusion:CUR may improve cardiac function in DCM rats by activating HIF-1α/VEGF signaling pathway to promote angiogenesis.关键词
