石决明配方颗粒的特异性PCR鉴别研究
Identification of Haliotidis Concha Formula Granules by Allele-Specific PCR
摘要
Abstract
Objective:To establish a specific polymerase chain reaction(PCR)method for the identification of Haliotidis Concha(Haliotis discus hannai)and its counterfeits,to make it suitable for raw materials of medicinal materials,freeze-dried powder of stand-ard decoction and formula granule.Methods:According to the single nucleotide polymorphism(SNP)sites of Haliotidis Concha(Haliotis discus hannai)and its counterfeits in cytopinase oxidase Ⅰ(COⅠ)gene sequence,the specific identification primers SJM-1F and SJM-1R of Haliotidis Concha(Haliotis discus hannai)were designed,and the main factors affecting PCR reaction were screened and op-timized,and the methodological investigation and sequencing were conducted.Results:At an annealing temperature of 59℃,26 cycles,the target bands of Haliotidis Concha(Haliotis discus hannai),standard decoction freeze-dried powder,formula granules were amplified at the same location about 123 bp,there was no interference in the corresponding position of the mixed counterfeit product and the blank control product.Conclusion:The established specific PCR method of Haliotidis Concha(Haliotis discus hannai)can effectively identify authentic and mixed counterfeit products and is not offected by raw materials and extraction and processing factors.It provides technical support for the quality control of the production,quality monitoring and market circulation of Haliotidis Concha(Haliotis discus hannai)formula granules.关键词
