Effects of Chlorogenic Acid on Autophagy and Inflammatory Response of ox-LDL-Induced Foam Cells by Regulating AMPK/SIRT1 Pathway

Abstract
Objective:To investigate the regulatory effects of chlorogenic acid on autophagy and inflammation in ox-LDL-induced foam cells through AMPK/SIRT1 signaling pathway.Methods:RAW264.7 macrophages were divided into blank control group,model group,chlorogenic acid(low,medium and high concentration)groups,chlorogenic acid high concentration with Compound C group.The degree of foam cells was observed by oil red O staining.The effect of chlorogenic acid on the proliferation of bubble cells was observed by MTT method.The autophagosomes were observed by transmission electron microscopy.The contents of TNF-α,IL-6 and IL-10 were detected by ELISA.The mRNA expressions of AMPK,SIRT1,Beclin1 and P62 were detected by fluorescence quantitative PCR.The protein expressions of SIRT1,ATG16L1 and P62 were detected by immunofluorescence labeling.Results:Oil red O staining showed that foam cells were successfully prepared after ox-LDL treatment.Compared with the model group,the number of autophagosomes in-creased,the secretion of TNF-α and IL-6 decreased,the secretion of IL-10 increased,the mRNA expressions of AMPK,SIRT1,Beclin1 and the protein expressions of SIRT1,ATG16L1 increased,and the mRNA and protein expressions of P62 decreased in each chlorogenic acid concentration group.Compound C could reduce the effect of chlorogenic acid.Conclusion:Chlorogenic acid can induce autophagy of bubble cells by regulating AMPK/SIRT1 pathway,down-regulate the overexpression of TNF-α and IL-6,and up-regulate the expression of IL-10,which may be one of the anti-AS mechanisms of chlorogenic acid.关键词
