摘要

Abstract

Objective:To observe the effect of acupotomy on cartilage degeneration and miR-107/Caspase-1/chondrocyte pyrop-tosis signal axis in knee osteoarthritis(KOA)rabbits.Methods:Twenty-four 6-month-old male New Zealand rabbits were randomly divided into blank group,model group and acupotomy group,with 8 rabbits in each group.In the model group and the acupotomy group,the classical Videman method was used to establish the rabbit KOA model.The blank group and the model group were not in-tervened.In the acupotomy group,cord-like or nodular structures around the quadriceps tendon and medial/lateral collateral liga-ments of the left knee were identified as acupotomy entry points.Acupotomy treatment was performed once weekly for 4 consecu-tive weeks.After the intervention,the wear of cartilage surface of the medial and lateral femoral condyle of the rabbit knee joint was observed.The pathological morphology of cartilage tissue was observed by hematoxylin and eosin(HE)staining and safranin O-fast green staining,and the pyroptosis of chondrocytes was observed by transmission electron microscope.Western blot was used to detect the protein expressions of Caspase-1,interleukin-1β(IL-1β),interleukin-18(IL-18)and Collagen Ⅱ in knee cartilage tissue.qPCR was used to detect the expression of miR-107,and Caspase-1,IL-1β,IL-18,Collagen Ⅱ mRNA.Results:Compared with the blank group,the knee joint cartilage surface of rabbits in the model group shows severe damage,the safranin staining of the cartilage ma-trix decreases,the degeneration of cartilage tissue is obvious,and obvious chondrocyte pyroptosis can be observed under the electron microscope.In the cartilage tissue of the model group,the expression levels of Caspase-1,IL-1β,IL-18(both protein and mRNA)are all increased(P<0.05),while the expression levels of Collagen Ⅱ(both protein and mRNA)and miR-107 are all decreased(P<0.05).Compared with the model group,the knee joint cartilage surface of rabbits in the acupotomy group is relatively smooth,the safranin staining of the cartilage matrix decreases slightly,the degeneration of cartilage tissue is alleviated,and the phenomenon of chondro-cyte pyroptosis is improved.In the cartilage tissue of the acupotomy group,the expression levels of Caspase-1,IL-1β,IL-18(both protein and mRNA)are all decreased(P<0.05),while the expression levels of Collagen Ⅱ(both protein and mRNA)and miR-107 are all increased(P<0.05).Conclusion:Acupotomy can delay the degeneration of cartilage tissue and regulate the expression of miR-107/Caspase-1/pyroptosis signal axis in cartilage tissue of rabbit KOA model.
