Bioinformatics analysis of the role of PAEP in the prognosis and immune regulation of hepatocellular carcinoma

Abstract
Objective:To systematically evaluate the expression profile of progestagen-associated endometrial protein(PAEP)in hepatocellular carcinoma(HCC),its prognostic value,and its potential role in regulating the tumor microenvironment(TME).Methods:RNA-seq data of HCC from TCGA database(371 HCC and 50 adjacent non-tumor tissues)were analyzed to assess the differential expression of PAEP and its association with prognosis.The relationship between PAEP expression and immune cell infiltration was evaluated based on 24 immune cell signature gene sets.Results:PAEP was significantly overexpressed in HCC,and elevated expression was strongly associated with poor prognosis(OS HR=1.82,95%CI:1.28-2.58;DSS HR=1.73,95%CI:1.10-2.70).The PAEP-based prognostic model exhibited good predictive performance(3-year AUC=0.720;5-year AUC=0.766).Immune microenvironment analysis revealed that PAEP expression was positively correlated with macrophage and Th2 cell infiltration,but negatively correlated with Th17 and CD8+T cell infiltration.Conclusion:PAEP may promote HCC progression and influence prognosis by modulating the tumor immune microenvironment and as such could become a potential target for combined immunotherapy.关键词
