Relationship between CD66b+tumor-associated neutrophils and epithelial-mesenchymal transition-related protein expression at the invasive front of colorectal adenocarcinoma cells and its clinicopathological significance

Abstract
Objective:Colorectal adenocarcinoma(COAD)is the most common subtype of colorectal cancer.Whether epithelial-mesenchymal transition(EMT)is involved in tumor-associated neutrophils(TAN)-mediated invasion of COAD remains unclear.This study aims to analyze the relationship between CD66b+TAN infiltration and the expression of EMT-related proteins(E-cadherin and β-catenin)at the invasive front of COAD cells,as well as their clinicopathological significance. Methods:A retrospective analysis was performed on 103 surgically resected,pathologically confirmed COAD specimens collected from Zhengzhou People's Hospital between 2019 and 2022.Immunohistochemistry using the EnVision method was conducted to evaluate the infiltration density of CD66b+TANs in COAD tissues[the mean number of CD66b+TANs per 10 high-power fields(HPFs)at the invasive front was used as the infiltration density].The membranous expression of E-cadherin and the aberrant nuclear expression of β-catenin in tumor cells at the invasive front were also assessed.Spearman's rank correlation analysis was performed to explore the relationship between CD66b+TAN density and EMT-related protein expression. Results:Among the 103 patients(60 males,43 females;aged 30-78 years,median 53 years),the infiltration density ranged from 0 to 54.0/10HPF,with an average of 18.67/10HPF.Patients with CD66b+TANs>18.67/10HPF were classified as the high-density group(n=50),and those with≤18.67/10HPF as the low-density group(n=53).At the invasive front,E-cadherin was lowly expressed in 37 cases(35.9%)and highly expressed in 66 cases(64.1%),while β-catenin showed high nuclear expression in 38 cases(36.9%)and low expression in 65 cases(63.1%).The high-density group had significantly higher rates of vascular invasion,lymph node metastasis,and advanced pTNM stage(III+IV)compared to the low-density group(P<0.05).There were no significant differences between groups in gender,age,tumor location,size,or differentiation(all P>0.05).CD66b+TAN infiltration density was significantly higher at the invasive front than in the tumor center(P<0.05).E-cadherin expression at the invasive front was significantly downregulated compared to the tumor center,whereas β-catenin nuclear expression was significantly upregulated(P<0.05).Spearman correlation analysis revealed that CD66b+TAN infiltration density was negatively correlated with E-cadherin expression(rs=-0.204,P=0.039)and positively correlated with aberrant nuclear expression of β-catenin(rs=0.264,P=0.007). Conclusion:The infiltration of CD66b+TANs at the invasive front of COAD cells is significantly associated with dysregulated expression of EMT-related proteins.It is hypothesized that CD66b+TANs at the invasive front may mediate abnormal expression of E-cadherin and β-catenin,thereby participating in EMT and promoting COAD progression.关键词
