夏枯草提取物、芦丁和槲皮素体外生物活性对比研究
Comparative Study on Biological Activities of Prunella vulgaris Extract,Rutin and Querce-tin in vitro

Abstract
[Objective]Extract storage and preservation of substances from traditional Chinese medicinal materials is an important direction in the field of agricultural product processing research.This study aimed to analyze the antioxidant capacity and antibacterial effect of Prunella vulgaris ex-tract,rutin and quercetin in vitro,and to provide a scientific basis for application of natural plant re-sources in preservation of agricultural products and development of functional foods.[Method]Four in vitro antioxidant assessment methods(DPPH,PTIO,ABTS free radical scavenging test,FRAP method)and Oxford cup antibacterial test were used to comprehensively analyze the differences in biological activities of the 3 substances.Vitamin C(VC)was used as the reference reagent in the in vitro antioxidant assessment,and the results were expressed as VC equivalent mg VCE/g.[Result]In the DPPH free radical scavenging assay,quercetin showed the highest scavenging effect(184.06 mg VCE/g),which was significantly higher than that of rutin(118.46 mg VCE/g)and Prunella vul-garis extract(6.69 mg VCE/g).There was no significant difference between quercetin and rutin(444.07 mg VCE/g and 469.37 mg VCE/g,respectively)in the PITO free radical scavenging assay,but both of them showed significantly higher effects than Prunella vulgaris extract(100.55 mg VCE/g).In the ABTS free radical scavenging assay,there was no significant difference between querce-tin(61.56 mg VCE/g)and rutin(58.11 mg VCE/g),but both of them showed significantly higher effexts than Prunella vulgaris(100.55 mg VCE/g).FRAP assay further confirmed that quercetin(1.06 mmol/L FeSO4)had the strongest iron reducing ability,which was not significantly different from that of rutin,but significantly higher than that of Prunella vulgaris extract(0.54 mmol/L Fe-SO4).Antibacterial test result showed that the inhibition zone diameters of rutin,quercetin and Prunella vulgaris extract against Staphylococcus aureus were 10.48 mm,9.98 mm and 10.30 mm,respectively,and those against Escherichia coli were 10.69 mm,9.82 mm and 10.41 mm,respec-tively.There was no significant difference among the 3 substances.[Conclusion]Quercetin has high antioxidant activity and the strongest iron ion reducing ability in vitro,but there was no significant difference in the antibacterial effect on Staphylococcus aureus and Escherichia coli among rutin,quercetin and Prunella vulgaris extract.关键词
