北部湾珊瑚礁区海绵中可培养细菌多样性及生物活性研究
Diversity and Bioactivities of Cultivable Bacteria Isolated from Sponges in the Coral Reef Reserve of the Beibu Gulf

Abstract
[Objective]To explore thecultivable microbial resources in sponges from the coral reef reserve of the Beibu Gulf and investigate their antibacterial and antitumor activities.[Methods]Fifteen sponge specimens were col-lected from the coral reef reserves of Xieyang Island and the Leizhou Peninsula in the Beibu Gulf.Twelve distinct bacte-rial isolation medium were employed for microbial isolation and cultivation using the dilution plating technique.Bacteri-al diversity was analyzed based on 16S rRNA gene sequencing.After primary antibacterial screening by the agar punch-ing method,the candidates showed antibacterial active were cultivated by fermentation.Then the secondary antibacteri-al screening and anti-prostate cancer active screening were applied on crude extracts from the above fermentation with MTT assay and filtering paper diffusion method.[Results]In total,235 cultivable bacteria representing 33 orders,61 families,and 99 genera were isolated from these 15 sponge specimens.Two species are proposed as putative novel spe-cies.The crude extracts of 21 species exhibited inhibitory activity against 10 pathogenic bacteria.The crude extracts of 8 species showed significant inhibitory activity against prostate cancer cells PC-3 and 22Rv1.[Conclusion]Sponges from the coral reef reserves of the Beibu Gulf contain richdiverse cultivable bacterial resources.Several isolated bacteri-al strains exhibit notable antibacterial and anti-prostate cancer activities,providing valuable strain resources to support the discovery and development of novel antibiotics and anticancer marine drugs.关键词
