基于转录组测序探讨光动力疗法对人牙周炎性组织mRNA表达谱的影响
Exploring the effect of photodynamic therapy on mRNA expression profile of human periodontic tissue based on RNA sequencing

Abstract
Objective:To explore the effect of photodynamic therapy(PDT)on gene expression in human periodontitis tissues based on RNA sequencing,analyze key differentially expressed genes(DEGs),and explore the potential mechanism of PDT.Methods:A total of 5 cases of healthy periodontal tissue,5 cases of periodontic tissue,and 5 cases of periodontic tissue treated with PDT were collected clinically,and RNA sequencing was performed.Bioinformatics analysis was used for hub gene screening and enrichment analysis.Another 15 cases of tissues for each group were collected and validated the sequencing results using RT-qPCR.Results:A total of 6 526 DEGs were detected between healthy periodontal tissue and periodontic tissue,and 1 209 DEGs were detected between periodontic tissue and periodontic tissue after PDT treatment,and 72 immune related differentially expressed genes were screened.GO enrichment analysis included biological processes such as leukocyte migration and bacterial de-fense response;KEGG pathway analysis mainly involved cytokine-cytokine receptor interactions,JAK-STAT signaling pathway,IL-17 signaling pathway,and other pathways.A PPI network was constructed and screen out 6 hub genes(S100A7,LCN2,LTF,DEFB4A,S100A12,S100A8).The RT-qPCR validation results were consistent with the sequencing results.Conclu-sion:The study determines the expression profile of mRNA in periodontitis tissue after PDT treatment,and identifies of relevant hub genes and functional analysis,can provide new ideas for studying the potential mechanism of PDT regulating of periodontitis tissue.关键词
