3D bioprinted GelMA/Bio-Oss/BMSCs composite scaffold study on repairing canine mandibular defects

Abstract
Objective:Evaluate the osteogenic efficacy of a 3D bioprinted composite scaffold composed of methacry-lated gelatin(GelMA)/bovine bone substitute(Bio-Oss)/bone marrow-derived mesenchymal stem cells(BMSCs)in repairing mandibular defects in Beagle dogs,providing a basis for developing novel bone regenerative materials.Methods:Mandibular defect models were established in the mandibles of 4 Beagle dogs(total of 24 defects).Based on the type of implanted materi-al,the dogs were randomly divided into four groups:Group A(implanted with GelMA/Bio-Oss/BMSCs composite scaffold),Group B(implanted with GelMA/Bio-Oss scaffold),Group C(blank control,only modeling without intervention),and Group D(positive control,implanted with Bio-Oss bone powder covered with Bio-Gide collagen membrane).At 4 and 12 weeks post-surgery,bone volume fraction(BV/TV)and bone mineral density(BMD)were analyzed using Micro-CT,and histological stai-ning(HE and Masson)was performed.Results:Micro-CT examination:Bone defect repair in Group A was significantly better than in Groups B and C(P<0.05).Compared with Group D,Group A exhibited faster degradation.Histological examination:Group A showed active new bone regeneration,and the bone quality was more mature compared to Group D.Histological quantitative analysis:Compared to Groups B and C,Group A had a significantly increased percentage of collagen fiber area(P<0.05).Conclusion:After implanting the GelMA/Bio-Oss/BMSCs composite scaffold into the mandibular defect of Bea-gle dogs,new bone tissue was stably formed,demonstrating good biocompatibility and osteogenic effects.关键词
