Construction Strategies and Advantageous Models of Perfused Vascular Systems in Cardiovascular Organoid Disease Modeling

Abstract
Cardiovascular organoids are models of ventricular like structural and functional characteristics constructed in vitro based on self-organizing developmental induction strategies,which are of great value in disease modeling,drug screening and toxicity assessment,regenerative medicine treatment,and so on.Aiming at the problem of nutrient delivery defects and insufficient physiologi-cal relevance caused by the lack of engineered perfusion system during the development of organoids,this paper focused on the charac-teristics of high metabolic demand of the heart,and clarified the necessity of dynamic fluid perfusion regulation by analyzing the inter-action mechanism of cell crosstalk and microenvironment in five types of cardiovascular disease models.This paper systematically reviewed the advantages and limitations of in vivo and in vitro modeling,and innovatively proposed three"perfusion vasculature"advantage models(vascularized organoid coculture,organ-on-chip technology,and microfluidic models),which provided a theoreti-cal framework for analyzing the regulatory mechanism of controllable perfusion on the maturity of cardiovascular organoids and promot-ing the clinical transformation of precise disease treatment.关键词
