Flow Cytometry-Based Dynamic Tracking Reveals the Migration Patterns of Peripheral T Lymphocytes in EAE Mice and Innovations in Dynamic Analysis Technology

Abstract
Objective The aim of this study was to study the proliferation,differentiation,and migration of T lymphocytes cells in spleen and lymph nodes of experimental autoimmune encephalomyelitis mice,and to explore the mechanism of peripheral T lympho-cytes in EAE mice.Methods The EAE model was established and its clinical score was recorded.The infiltration of nucleated cells in the spinal cord of EAE was observed by HE staining.The total numbers of splenocytes and lymph-node cells in four groups of mice(0,5,10 and 15 days after EAE induction)were counted using an abalone counting board.Combined with flow cytometry(FCM)at mul-tiple time points,the changes of the proportion and activation ratio of CD4+and CD8+T lymphocytes in spleen and lymph node of the four groups were analyzed.Results The spinal cord tissue of EAE mice was infiltrated by a large number of nucleated cells;the total number of spleen cells and lymph node cells increased gradually after EAE induction,but decreased sharply on the 15th day(P<0.005);the proportions of CD4+and CD8+T lymphocytes decreased progressively but rebounded on day 15(P<0.005),whereas their absolute counts rose gradually before dropping sharply on day 15(P<0.05);Furthermore,the activated proportion of T lymphocytes showed a significant increasing trend.Conclusions The differentiation,proliferation,and activation of peripheral T lymphocytes were accelerated from the induction of EAE to the initial stage of the disease,and the sharp decline in the peak stage of the disease might be related to the migration of pathogenic T lymphocytes to Spinal cord.This study,for the first time,revealed the sudden drop pattern of pathogenic T cell migration through dynamic flow cytometry technology,providing a new monitoring strategy for targeted therapy of mul-tiple sclerosis.关键词
