三碘甲状腺原氨酸通过激活Nrf2信号通路促进小鼠皮肤伤口愈合的作用研究
Triiodothyronine Promotion Skin Wound Healing via Activating of Nrf2 Signaling in Mice

Abstract
Objective This study aimed to explore whether triiodothyronine(T3)can promote skin wound healing in mice by activating Nrf2 signaling pathway.Methods The mice were randomly divided into four groups:the normal group,the control group,the T3 group,and the ML385(an Nrf2 inhibitor)+T3 group.A full-thickness skin defect model was created in all mouse groups except for the normal group.The wound healing progression was observed daily.The mice were killed respectively on post-injury days 3,7,and 14,with 8 mice in each group.Pathological change and formation of collagen fibers were observed by HE and Masson stain-ing respectively.The expression of caspase-3 was evaluated by immunohistochemistry staining.The contents of ROS,MDA,CAT,GSH,SOD,IL-6,and TNF-α were quantified using ELISA assay.The Nrf2 and HO-1 protein levels were determined by Western blot.Results On days 3 and 7 post-injury,wound healing rates in T3 group were higher than those in the control group and the ML385+T3 group(P<0.05).The T3 group showed superior improvement in pathological changes compared to the control group and the ML385+T3 group.On day 7 post-injury,the T3 group showed significantly higher levels of collagen fiber formation and caspase-3 expression compared to both the control and ML385+T3 groups(P<0.05).However,these levels were significantly lower in the T3 group than in both control groups by day 14 post-wound(P<0.05).Furthermore,on days 3 and 7 post-wound,the T3 group exhibited significantly lower levels of ROS,MDA,IL-6,and TNF-α,but higher levels of SOD,GSH,Nrf2,and HO-1 compared to both the control and ML385+T3 groups(P<0.05).Conclusions T3 may promote skin wound healing by activating the Nrf2 signaling path-way,through mechanisms involving enhanced antioxidant capacity,modulation of the inflammatory response,regulation of apoptosis,and facilitation of extracellular matrix remodeling.关键词
