Isolation,Identification,and Biological Characteristics of Protease-Producing Shewanella from the Intestinal Tract of Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂

Abstract
Shewanella,a class of Gram-negative proteobacteria ubiquitously distributed in natural environments,is frequently identified as an indicator of food spoilage.The Pearl Gentian grouper(Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂)is renowned for its delicious flavor,but its susceptibility to spoilage poses significant challenges for preser-vation.To elucidate the biological characteristics of intestinal Shewanella in this species,two strains,designated ZL01 and ZL07,were isolated from the intestinal contents and phylogenetically identified.Their 16S rDNA sequence similari-ties to S.seohaensis were 0.978 and 0.964,respectively.The results showed that both strains ZL01 and ZL07 exhibited broad adaptability to pH.The optimal growth pH was 7 for ZL01 and 6 for ZL07.ZL01 grew within a NaCl concentra-tion range of 0%to 7%,while ZL07 showed optimal growth without NaCl.The optimal growth temperatures for ZL01 and ZL07 were 30℃and 35℃,respectively.The results indicate that appropriately manipulating storage parameters,particularly temperature and NaCl concentration,can effectively suppress Shewanella proliferation,thereby extending the shelf life of Epinephelus fuscoguttatus♀×E.lanceolatus♂.This study provides a theoretical basis for the development of novel antimicrobial strategies.关键词
