摘要

Abstract

The Wnt and Notch signaling pathways,which play crucial roles in hair cell development,are also piv-otal in hair cell regeneration.To explore techniques for regenerating adult mouse cochlear hair cells,cochleostomy was employed.Recombinant adenovirus was microinjected into the scala media of adult mouse cochleae to infect supporting cells and modulate the activity of intracellular Wnt and Notch signaling pathways.Results showed that adenovirus-mediated alterations in both Wnt and Notch signaling pathways induced regeneration of cochlear hair cells in adult mice.Compared to modulating a single pathway alone,simultaneously altering the activity of both Wnt and Notch pathways demonstrated significantly greater effects on hair cell regeneration and supporting cell proliferation.Further analysis con-firmed that supporting cells subjected to dual-pathway regulation could proliferate and regenerate hair cells expressing marker genes characteristic of mature hair cells.This technique offers an effective approach for investigating hearing im-pairments resulting from cochlear hair cell damage.
