Isolation,culture and identification of primary hepatocytes from Schizothorax o'connori

Abstract
To establish the in vitro isolation and culture of primary hepatocytes from Schizothorax o'connori and explore their biological characteristics,the liver tissue of S.o'connori was digested by collagenase digestion,and the primary hepatocytes were obtained by density gradient centrifugation.The cell morphology was observed.The growth cycle of hepatocytes was detected by CCK-8 kit and the growth curve was plotted.The isolated and purified primary hepatocytes were identified by periodic acid-Schiff staining(PAS)and cytokeratin-18(CK-18)immuno-fluorescence staining.The results showed that the morphology of primary hepatocytes of S.o'connori were intact and adhered well.However,the 4th-generation cultured cells failed to adhere and grow after digestion.The fluo-rescence staining of PAS and CK-18 was positive,and the purity of hepatocytes was above 90%,indicating suc-cessful isolation of primary hepatocytes of S.o'connori.关键词
