基于数据挖掘和网络药理学探究含当归复方治疗肾性贫血的用药规律及作用机制
Medication regularity and mechanism of action research of Angelica sinensis compound in treating renal anemia based on data mining and network pharmacology

Abstract
Objective Based on data mining,the medication rules of Angelica sinensis compound in the treatment of renal anemia were analyzed,and the mechanism of its action in improving renal anemia was explored by network pharmacology.Methods The relevant literatures on the treatment of renal anemia with Angelica sinensis compound were screened from China Academic Journal Full-text Database,Wanfang Data Knowledge Service Platform,VIP Biomedical Database,Chinese Biomedical Literature Database and PubMed database.The frequency of traditional Chinese medicine(TCM)use,nature,taste and meridian of all the compounds were analyzed.Association rules and cluster analysis were used to determine the drug compatibility of the compound and the core drug combination.Network pharmacology was used to analyze the related mechanism of the core drug combination in improving renal anemia,and molecular docking technology was used to verify.Results A total of 211 prescriptions that met the inclusion criteria were screened,involving 156 Chinese herbs,with a total frequency of medication of 2498 times.The most commonly used category was tonics,and the majority of them were warm in nature,sweet in taste and spleen meridian.The core drug combination had 26 components and 211 potential targets,such as tumor protein p53(TP53),protein kinase B α(AKT1),Heat Shock Protein 90 Alpha Family Class A Member 1(HSP90AA1),estrogen receptor 1(ESR1),and Proto-Oncogene Tyrosine-ProteinKinase Src(SRC),etc.Gene ontology(GO)functional analysis suggested that its mechanism of action was involved in biological processes such as hormone regulation,oxidative stress,and apoptosis.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis suggested that it was related to phosphatidylinositol 3-kinase-protein kinase B(PI3K-Akt)signaling pathway,advanced glycation end-products-receptor for AGE(AGE-RAGE)axis,tumor necrosis factor(TNF)signaling pathway and other pathways.Molecular docking validation revealed that quercetin exhibits strong binding affinity towards AKT1,while kaempferol demonstrates favorable binding activity with SRC and ESR1.Conclusion The core drug combination of Angelica sinensis,Astragalus membranaceus and Atractylodes macrocephala can promote hematopoiesis,regulate iron metabolism,inhibit inflammatory response and apoptosis through multi-components,multi-targets and multi-pathways,which provides a basis for the mechanism research of TCM in the treatment of renal anemia.关键词
