摘要

Abstract

Objective To explore the mechanism of Sanqi Shengban Decoction in treating primary immune thrombocytopenia(ITP)using network pharmacology,and to provide basis and reference for future research.Methods To obtain the active ingredients and related targets of Sanqi Shengban Decoction through the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),and to use UniProt database to integrate and summarize the target genes of action,to apply GeneCard,PharmGkb,TTD,OMIM,DrugBank database to obtain ITP disease targets and extract drug disease intersection targets.To construct a drug-component-disease target network diagram using Cytoscape 3.10.2 software;To construct a protein-protein interaction(PPI)network using STRING database and to analyze it using Cytoscape 3.10.2 software;To apply the Microbioinformatics data platform to perform Gene Ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis on intersecting targets.To use software such as PyMOL and AutoDock Tools for molecular docking.Results A total of 158 active ingredients,199 potential targets,1419 ITP disease targets,and 100 intersection targets between Sanqi Shengban Decoction and diseases were selected through screening.And key active ingredients such as quercetin,kaempferol,β-sitosterol,naringenin,7-methoxy-2-methylisoflavone,mangiferin,sitosterol,Medi rosewood extract,isorhamnosin,and lignan have been screened out.The GO enrichment analysis results showed that the key targets involved a total of 1503 biological processes,58 cell compositions,and 134 molecular functions;KEGG enrichment analysis results showed that a total of 179 pathways were involved in key targets,including cancer pathway,chemical carcinogenesis receptor activation,cancer transcription disorder,lipids and atherosclerosis,fluid shear stress and atherosclerosis signal pathway,hypoxia inducible factor-1(HIF-1)signal pathway,nuclear factor κB(NF-κB)signal pathway,etc.The molecular docking results between the core target and the core active ingredient are good.Conclusion Sanqi Shengban Decoction may be related to cancer pathway,chemical carcinogenesis receptor activation,cancer transcription disorder,lipid and atherosclerosis,fluid shear stress and atherosclerosis signal pathway,HIF-1 signal pathway,NF-κB signal pathway and other pathways in the treatment of ITP.Quercetin,kaempferol,β-sitosterol,naringenin,7-methoxy-2-methylisoflavone,formononetin,sitosterol,melatonin,isorhamnetin,and xylanone may be the key active ingredients in the treatment of ITP.
