Abstract

To explore the molecular mechanisms of chemical hybridizing agent SQ-1-induced pollen abor-tion in wheat,this study was conducted using the wheat cultivar Xinong 1376 as experimental material.The RNA-seq technology was employed to perform transcriptome sequencing of wheat florets treated with 5 kg·hm-2 SQ-1(PHYMS-1376)and water(control,CK-1376)at 8 h,24 h and 5 d.Differentially expressed genes(DEGs)were screened,followed by GO functional annotation,KEGG pathway analysis,and expression pattern investigation.The results showed that a total of 2 827 DEGs were identified,including 1 799 significantly upreg-ulated genes and 1 028 significantly downregulated genes.GO enrichment analysis indicated that the DEGs were significantly enriched in the biological processes such as photosynthesis and DNA replication,the cellular com-ponents such as thylakoid membrane and cell wall,and the molecular functions mainly related to electron trans-fer activity and ATPase activity.KEGG pathway analysis indicated that the DEGs were mainly participated in the pathways such as starch and sucrose metabolism,glutathione metabolism and phenylpropanoid biosynthesis.The expression patterns of 16 DEGs obtained by qRT-PCR analysis were highly consistent with the transcriptome a-nalysis results(R2=0.985 6).In conclusion,SQ-1 might induce male sterility by disrupting key pathways such as reactive oxygen species metabolism,carbon source allocation and energy supply,ultimately leading to im-paired synthesis of pollen development-related substances.These findings could provide a theoretical basis for further exploration of the molecular mechanisms of SQ-1 and its application in wheat heterosis utilization.
