好氧堆肥对牛粪中养分含量及抗生素抗性基因丰度的影响
Effects of Aerobic Composting on Nutrient Content and Abundance of Antibiotic Resistance Genes in Cow Manure

Abstract
To investigate the effects of aerobic compost on nutrient content and the abundance of antibiot-ic resistance genes(ARGs)in cow manure,the cow manure and sorghum straw were mixed at a volume ratio of 7∶3 in this study,and aerobic composting was carried out under natural conditions.The samples before composting were CK,and those after composting were CP.The temperature during composting was monitored,the seed germination index after treated with the sample extracts and the nutrient contents before and after composting were analyzed,the types and abundance of ARGs were detected by high-throughput qPCR chip,and the correlation between the abundance of ARGs and nutrient content was analyzed.The results showed that the high temperature period during aerobic composting was 9 days,and the highest temperature was 72℃.The germination index of cucumber and radish seeds treated with CP extract was 117.91%and 97.82%,which was 98.23%and 73.87%higher than that of CK.Compared with CK,the pH value and contents of total nitrogen,total phosphorus and total potassium of CP increased by 2.33%,8.04%,20.18%and 59.98%,and the organ-ic matter content significantly decreased by 10.43%.In CK,a total of 13 major categories of ARGs were detec-ted which were aminoglycoside antibiotic,tetracycline antibiotic,macrolide-lincophthalide-streptococcin B(MLSB),lincoamides(multidrugs),diaminopyrimidine antibiotic,phenicol antibiotic,beta-lactamase,fluo-roquinolone antibiotic,glycopeptide antibiotic,insertional,sulfonamide antibiotic,peptide antibiotic and transposases and a large class of mobile gene elements(MGEs).In CP,12 major classes of ARGs(except peptide antibiotics)and one major class of MGEs were detected.There were 102 ARGs from CK detected,and 80 genes detected from CP with the detection rate decreased by 21.57%.The top three ARGs in CK and CP were aminoglycoside antibiotic,tetracycline antibiotic and MLSB with the detected numbers of 26 and 23,17 and 13,12 and 9,respectively.The removal rate of CP for aadA17 gene and IncI1_repI1 and ISCR1 mobile genetic elements was 100%.Correlation analysis showed that total nitrogen and total phosphorus had significant effects on the absolute quantitative abundance of floR,sul2,sul1,qacF_H,tnpA-3,tetX,IS6100,IS1247,ISEcp1 and ISCR1,while organic matter had significant effects on the absolute quantitative abundance of IncI1_repI1 and APH(6)-Id.The abundance of aadA2 and aadA17 was significantly affected by pH value and total potassium.In conclusion,aerobic composting could improve the seed germination index and nutrient content of cow manure,reduce the absolute quantitative abundance of ARGs and MGEs in cow manure,improving the quality of cow manure resource utilization and reducing the potential risk of cow manure resource utilization.关键词
