Correlation of serum circular RNA MACF1 and circular RNA Ube3a expressions with myocardial injury in patients with acute myocardial infarction

Abstract
Objective To explore the correlation of serum circular RNA MACF1(circMACF1)and circular RNA Ube3a(circUbe3a)expressions with myocardial injury in patients with acute myocardial infarction(AMI).Methods A total of 80 patients admitted to the Second Affiliated Hospital of Zhejiang University School of Medicine,Linping Campus from May 2022 to May 2024 and diagnosed with AMI were selected as the research subjects(AMI group).They were treated with thrombolysis and received percutaneous coronary intervention(PCI).Another 90 healthy individuals who underwent physical examinations were selected as the control group.General information of the subjects was collected.qRT-PCR was applied to determine the expression levels of serum circMACF1 and circUbe3a.AMI patients were grouped into the cardiac function grade Ⅰ group(41 cases),cardiac function grade Ⅱ group(19 cases),and cardiac function grade Ⅲ group(20 cases)based on the Killip classification of cardiac function.Fully automated biochemical analyzer was applied to detect the level of creatine kinase isoenzyme(CK-MB).Fully automatic electrochemiluminescence instrument was applied to detect the levels of procalcitonin(PCT),brain natriuretic peptide(BNP),cardiac troponin I(cTnI),and myoglobin I(Myo).The correlation of serum circMACF1 and circUbe3a with cardiac function and myocardial injury markers in AMI patients was analyzed using Pearson method.Results The serum level of circMACF1 in the AMI group decreased compared to the control group,while the serum circUbe3a,BNP,PCT,Myo,cTnⅠ,and CK-MB levels increased(P<0.05).After treatment,serum circMACF1 and left ventricular ejection fraction in AMI group were higher than before treatment,while serum circUbe3a,BNP,PCT,Myo,cTnI,and CK-MB were lower than before treatment(P<0.05).In patients with cardiac function gradesⅠ,Ⅱ,and Ⅲ,the serum expression level of circMACF1 gradually decreased,while the expression level of circUbe3a gradually increased(P<0.05).The serum circMACF1 level in AMI patients was negatively correlated with cardiac function,BNP,PCT,Myo,cTnⅠ,and CK-MB levels,while circUbe3a level was positively correlated with cardiac function and the above biomarkers(P<0.05).Conclusion In AMI patients,serum expression level of circMACF1 is decreased,while circUbe3a level is increased,both of which are related to myocardial injury.关键词
