Abstract

Objective To investigate the therapeutic effects and potential mechanism of Qingzhuan Dark tea extract(QDTE)on ul-cerative colitis(UC)in mice induced by dextran sulfate sodium(DSS).Methods A UC model was established in C57BL/6 mice by 3%DSS,and the mice were intervened with 200mg/(kg·d)and 400mg/(kg·d)of QDTE,respectively.The therapeutic effect of QDTE was evaluated by monitoring body weight changes,disease activity index(DAI),and colon length.The levels of inflammatory factors(IL-6,TNF-α)in serum and colon tissue were detected by enzyme-linked immunosorbent assay(ELISA),and the expression changes of zonula occlu-dens-1(ZO-1)and occludin were analyzed by immunohistochemistry.Results Compared with the Model group,QDTE intervention signifi-cantly alleviated the weight loss of UC mice(P<0.05),reduced DAI scores(P<0.05),and increased colon length(P<0.05).In addi-tion,QDTE significantly inhibited the release of pro-inflammatory factors IL-6 and TNF-α in the colon and serum(P<0.05),and upregu-lated the expression of ZO-1 and Occludin in the intestinal mucosa(P<0.05).Conclusion QDTE effectively alleviates the symptoms of DSS-induced UC in mice by inhibiting inflammatory responses and enhancing intestinal barrier function,indicating its potential application value as a functional food or adjuvant therapy for UC.
