Abstract
Objective To explore the effect of Baiji Zhishi Decoction on indomethacin-induced acute gastric ulcer in rats and its mechanism of action.Methods Forty-eight healthy male SD rats were randomly divided into 6 groups:control group,model group(60mg/kg indomethacin),positive control group(indomethacin+40 mg/kg omeprazole),low,medium and high dose Baiji Zhishi Decoction groups(indomethacin+10,20,40 mg/kg Baiji Zhishi Decoction).The rats were given the drugs once a day by gavage for 15 consecutive days.The model was established 8 hours after the last gavage,and the samples were collected 6 hours after the model was established.The contents of SOD,EGF,PGE2,MDA,NO,TNF-α and IL-6 in serum of each group were measured,and the gastric ulcer index of rats was compared.The pathological observation was conducted by HE staining.The apoptosis of gastric mucosal epithelial cells were observed by TUNEL method.The content of pepsin was activity.The expression of PI3 K,AKT,Caspase3,BAX and Bcl2 proteins in gastric tissue of each group of rats were determined by Western blot.Results Compared with the control group,the contents of EGF,PGE2,NO and SOD in the model group were significantly decreased(P<0.001),while the contents of IL-6,TNF-α,MDA determined and pepsin activity were significantly in-creased(P<0.01).The number of apoptotic cells in the gastric mucosa was significantly increased,and the bleeding of the gastric mucosa was severe.The expression of PI3K,AKT and Bcl2 proteins was significantly decreased(P<0.001),while the expression of BAX and Caspase3 proteins was significantly increased(P<0.01).Compared with the model group,the contents of EGF,PGE2,NO and SOD in the Baiji Zhishi Decoction groups were significantly increased(P<0.05),while the contents of IL-6,TNF-α,MDA determined and pepsin ac-tivity were significantly decreased(P<0.05).The linear bleeding of the gastric mucosa was improved,the number of apoptotic cells in the gastric mucosal epithelium was reduced,and the expression of PI3K,AKT and Bcl2 proteins was significantly increased(P<0.05),while the expression of BAX and Caspase3 proteins was significantly decreased(P<0.05).Conclusion Baiji Zhishi Decoction may exert a pro-tective effect on acute gastric ulcer in rats by activating the PI3K/Akt pathway,inhibiting apoptosis,promoting cell proliferation and reducing the release of inflammatory factors.关键词
