摘要

Abstract

Objective To investigate the protective effect of Celastrol(CEL)on the cerebral cortex of diabetic rats.Methods A rat model of diabetes mellitus(DM)was established by combining a high-fat diet with an intraperitoneal injection of streptozotocin(STZ,35mg/kg).The treatment group was administered CEL(4mg/kg/day)orally for 12 weeks.The conditioned fear experiment was used to as-sess the rats' learning and memory abilities.Nissl staining was performed to examine the morphology and number of cortical neurons.Immu-nofluorescence was used to detect the expression level of p-NF-κB in the cerebral cortex.Western blot analysis was conducted to measure the expression of p38AMPK and STAT3 proteins.Results Compared with the DM group,the CEL-treated rats showed more orderly arrangement,regular morphology of cortical neurons,with no significant cell vacuolation.Additionally,the number of Nissl bodies was increased,p-NF-κB expression was reduced,and the expression of p38AMPK and STAT3 proteins were downregulated.Conclusion CEL exerts a protective effect on the cerebral cortex of DM rats and improves learning and memory abilities,which may be related to the p38AMPK/STAT3 signaling path-way.
