Abstract

Objective To investigate the role and mechanism of metformin(Met)in regulating macrophage polarization and inflam-matory responses.Methods Using lipopolysaccharide(LPS)-stimulated RAW 264.7 macrophages as a model,cells were divided into six groups:normal control group(Con),the LPS group,the LPS+metformin group(2mmol/L),the LPS+metformin group(10mmol/L),the LPS+metformin group(50mmol/L),and the LPS+metformin group(100mmol/L).Cell morphology was observed under a microscope and cell viability was determined by CCK8.The secretion of IL-1β,IL-6 and TNF-α were detected using ELISA.The protein expression levels of Nur77 and NF-κB were detected by Western blot.Results The microscopic results showed that compared with the normal control group,LPS stimulation activated macrophages and promoted polarization toward the M1 phenotype.Metformin intervention ameliorated LPS-induced mac-rophage damage and restored normal cell morphology.The CCK8 results showed that treatment with 50mmol/L metformin for 12 hours yiel-ded the highest cell viability.ELISA results showed that compared with the normal control group,LPS significantly increased the levels of IL-1β,IL-6,and TNF-α(all P<0.0001),which were most markedly reduced by the addition of 50mmol/L metformin(P<0.0001).Western blot results showed that compared with the normal control group,LPS significantly upregulated NF-κB protein expression(P<0.0001)and downregulated Nur77 expression(P<0.05).In contrast,compared with the LPS group,metformin treatment significantly decreased NF-κB expression(P<0.001)and increased Nur77 expression(P<0.001).Conclusion Metformin can enhance the anti-inflammatory ability of macrophages and affect macrophage polarization by regulating Nur77,thereby inhibiting inflammatory responses.These findings provide new ideas for targeted treatment of various inflammatory diseases.
