Abstract

To explore the distribution characteristics and regulatory mechanisms of melanin synthesis-related sub-stances in Minxian Black Fur sheep,in this study,6 Minxian Black Fur Sheep were taken as the research objects,and 6 Small-tailed Han sheep were used as the control.Enzyme linked immunoabsorbent assay(ELISA)was employed to de-tect tyrosine(Try),tyrosinase(TRY),tyrosinase-related protein 1(TYRP1),and tyrosinase-related protein 2(DCT)in 14 parts of the experimental sheep,including the heart,liver,spleen,lung,kidney,tongue,rumen,abomasum,duode-num,rectum,skin,muscle,subcutaneous fat,and rib.Subsequently,3 sheep were randomly selected from each group,and the expression levels of TYR,TYRP1,DCT,PMEL,OCA2,and MC1R genes in their skin tissues were detected by RT-qPCR.The regulatory mechanism of melanin deposition in Minxian Black Fur sheep was comprehensively ana-lyzed.The results revealed that Try,TRY,TYRP1,and DCT were present in all the tested parts of the experimental sheep,and their concentration/enzyme activity in each tissue of Minxian Black Fur sheep were higher than those of Small-Tailed Han sheep(P<0.05).The concentration of Tyr was the highest in the liver of both Minxian Black Fur sheep and Small-tailed Han sheep,followed by the heart,and the lowest in the skin tissue.The activity of TYR was the high-est in the liver tissue of both breeds,followed by the heart and rumen,and the enzyme activity was lower in motor or-gans such as the skin,subcutaneous fat,muscle,and rib.The concentration of TYRP1 was highest in the liver tissues of the two varieties,significantly higher than other tissues(P<0.05),followed by the heart.The activity of DCT was rela-tively high in the liver and heart of both Minxian Black Fur sheep and Small-tailed Han sheep.The expression levels of TYR,TYRP1,DCT,PMEL,OCA2,and MC1R genes in the skin tissue of Minxian Black Fur sheep were extremely significantly higher than those of Small-tailed Han sheep(P<0.01).The relative expression levels of TYR,TYRP1,DCT,PMEL,OCA2,and MC1R genes were extremely significantly positively correlated with the activities of TYR and DCT(P<0.01),and significantly positively correlated with the concentration of Tyr(P<0.05).The relative expression levels of TYRP1 and OCA2 genes were only significantly positively correlated with the concentration of TYRP1(P<0.05),and the correlation with other indicators was not significant(P>0.05).In conclusion,melanin synthesis was found in all tissues of the sheep,with the highest concentration in liver.The regulatory effects of melanin deposition-related genes TYR,TYRP1,DCT,PMEL,OCA2,and MC1R lead to differences in melanin synthesis-related substances,which are important reasons for the different skin colors of Minxian Black Fur sheep and Small-tailed Han sheep.
