Abstract

The animal gut microbiome is a dynamic equilibrium symbiosis system established through host-microbial interactions,and its community structure and function play important biological roles in host energy metabolism,im-mune regulation,ecological adaptation,and health assessment.This study focused on the Qinling takin(Budorcas taxi-color bedfordi),an endangered species in China.The PacBio's 16S rRNA full-length sequencing was employed to re-veal the gut microbiota composition of 14 takins(4 in infant group,5 in adult group,and 5 in old group)from the Shaanxi Rare Wildlife Rescue Base.The results showed that there was no significant difference in the Alpha diversity index of the gut microbiota among different age groups of takins(P>0.05).However,NMDS analysis based on weighted_unifrac distance showed significant differences in the gut microbiota between groups(P=0.01).Under dietary homogeneity conditions,Firmicutes,Bacteroidetes,and Proteobacteria,were dominant phyla in the gut microbia of ta-kin,and their relative abundances exhibited variations across age groups.The abundance of Firmicutes in the adult group was higher than that of the old group and infant group(P<0.05).The abundance of Bacteroidetes was the highest in the infant group,and the abundance of Proteobacteria was the highest in the old group.At the genus level,Bacteroi-des was significantly enriched in the infant group,and the abundance of Bibersteinia was higher in the old group.Li-near discriminant analysis(LEfSe)analysis identified markers of characteristic bacterial genera in different age groups,among them,the adult group had the most(14),the infant group had 12,and the old group had the least(6).PICRUSt functional prediction showed that the gut microbiota of takin involved in pathways such as global and overview maps,carbohydrate metabolism,amino acid metabolism,membrane transport,and translation.At level 2,there were five meta-bolic pathways with significant differences between the old group and the infant group(P<0.001).This study explored the composition and potential functions of gut microbiota in takin of different ages,provids the important reference for formulating protection and management strategies for takins.
