Abstract

In order to screen miRNA related to ovarian development in Bactrian camels,6 healthy non-pregnant Bac-trian camels with similar age and body weight were selected as the study subjects and divided into 2 groups(reprodu-tive season group and non-reprodutive season group)according to the test requirements.Transcriptome sequencing was used to analyze miRNA in ovarian tissues of Bactrian camel,and the differentially expressed miRNAs were screened.The GO function and KEGG pathway enrichment of differentially expressed genes were analyzed,and mi-RNAs related to ovarian development were screened.Four differentially expressed miRNAs were randomly selected for verification.The results showed that 741 miRNAs were expressed in both the reproductive and non-reproductive sea-sons,58 miRNAs were expressed specifically in the reproductive season,and 60 miRNAs were expressed specifically in the non-reproductive season.47 differentially expressed miRNAs were identified by screening.Compared with non-reproductive season,26 miRNAs were significantly up-regulated and 21 miRNAs were down-regulated in the ovary during reproductive season.KEGG enrichment analysis showed that miRNA target genes were mainly enriched in the signaling pathways related to ovarian development,such as MAPK,PI3K-Akt,AMPK,Hippo,Rap1,p53,and Wnt.Four differentially expressed miRNAs(miR-10b,miR-361,miR-196a,and miR-345-5p)participating in the above signaling pathways were selected for qRT-PCR verification.The results showed that miR-10b and miR-361 were sig-nificantly up-regulated(P<0.05),miR-196a was significantly up-regulated(P<0.01),and miR-345-5p was signifi-cantly down-regulated(P<0.01)in the ovaries of female camels during the reproductive season,which were consistent with those of miRNA sequencing.These results suggest that miRNA plays an important role in ovarian development of Bactrian camels.
