多胎型和单胎型绵羊间情期卵巢发育关键lncRNA的筛选与鉴定
Screening and Identification of Key lncRNAs for Ovarian Development During the Anoestrus of Multipe and Single Fetal Sheep

Abstract
This study aimed to explore the molecular regulatory mechanism of lncRNA(Long non-coding RNA)in the ovarian development of different breeds of sheep,identify genes related to sheep reproduction,and provide reference for improving production efficiency.In this study,the expression profiles of non-coding RNA in ovarian tissue of multi-fetal(Small-tailed Han sheep)and single-fetal(Ujumuqin sheep)sheep were constructed.DE lncRNAs,DE miRNAs and DE mRNAs were screened through high-throughput sequencing analysis,and their target genes were pre-dicted and GO and KEGG enrichment analysis were performed.The ceRNA(lncRNA-miRNA-mRNA)regulatory net-work was further constructed.The results showed that a total of 1 579 lncRNAs,175 miRNAs and 3 095 mRNAs diffe-rentially expressed in the ovarian tissues of Ujumuqin sheep and Small-tailed Han sheep were screened.The enrichment analysis of differentially expressed lncRNAs target genes was annotated to reproductive signaling pathways such as ste-roid biosynthesis,ketone body synthesis and decomposition,and histidine metabolism,and then two key lncRNAs regu-lating ovine ovarian development were screened in combination with ceRNA network.lncRNA MSTRG.26091 might regulate the expression of target gene SDR42E2 by binding to bta-miR-331-3p,and lncRNA MSTRG.18078 might regulate the expression of target gene BCAS1 by binding to bta-miR-197,both of which were involved in ste-roid biosynthesis signaling pathway and affected ovarian development in sheep.Real-time fluorescence quantifica-tion(qRT-PCR)results were consistent with the trend of high-throughput sequencing results,indicating the reliabi-lity of sequencing data.关键词
