葡萄籽原花青素对马精液低温和冷冻保存质量的影响
Effect of Grape Seed Anthocyanin on the Quality of Horse Semen under Low-temperature and Frozen Preservation

Abstract
To investigate the effect of grape seed procyanidin(GSP)on the quality of horse semen and its antioxidant capacity after low temperature and frozen storage,this experiment was conducted by adding different concentrations of GSP(0,0.02,0.04,0.06,0.08 g/L)to the horse semen diluent,and then storing them in a refrigerator at 4℃and liquid nitrogen for low temperature and frozen storage,respectively.The sperm survival rate,plasma membrane integrity,ac-rosome integrity,average path velocity(VAP),straight velocity(VSL),and curvilinear velocity(VCL)were measured after 1-7 days of low temperature storage as well as after freezing and thawing.The glutathione peroxidase(GSH-Px),total antioxidant capacity(T-AOC),and catalase(CAT)were also measured to evaluate effects of GSP on the quality of horse semen after low temperature and frozen storage.The results showed that compared with the control group,adding an appropriate amount of GSP to the base solution can improve sperm survival rate,plasma membrane integrity rate,ac-rosome integrity rate,VAP,VCL,VSL,T-AOC,CAT,and GSH-Px activity(P<0.05)within 7 days of low-temperature storage,with the best preservation effect observed in the 0.06 g/L concentration group.During the freezing process,com-pared with the control group,adding 0.02-0.06 g/L GSP to the freezing solution had an increasing effect on sperm sur-vival rate,membrane integrity rate,acrosome integrity rate,VAP,VCL,and VSL(P<0.05),and the activities of T-AOC,CAT,and GSH-Px in semen were the highest(P<0.01)at administrated of 0.06 g/L GSP.Under the conditions of this experiment,GSP can improve the sperm survival rate,plasma membrane integrity,acrosome integrity,motility para-meters and antioxidant enzyme activity of horse sperm after low-temperature and frozen storage,effectively improving the quality of low-temperature and frozen storage of sperm,and the optimal addition concentration is 0.06 g/L.关键词
