摘要

Abstract

To further explore the maternal genetic diversity,population genetic structure,and genetic background of Gannan yaks at the molecular level,the present study determined the mtDNA D-loop region sequences of 34 Gannan yaks.Based on this,23 published Gannan yak mtDNA D-loop region sequences were downloaded from GenBank,and a total of 57 Gannan yak mtDNA D-loop region sequences were comprehensively analyzed using bioinformatics software such as BioEdit v7.2.5,DnaSP 5.10.01,and Arlequin 3.11.The results showed that a total of 4 single polymorphic sites and 33 parsimonious informative sites were detected in the comparative analysis of 57 Gannan yak mtDNA D-loop se-quences.A total of 19 haplotypes were identified based on nucleotide variations among sequences.Genetic diversity analysis revealed that compared to wild yaks and 17 other domestic yak breeds in China,Gannan yaks have higher haplo-type diversity(0.922±0.017)and nucleotide diversity(0.016±0.008),indicating a higher level of maternal genetic diver-sity.In phylogenetic analysis,19 haplotypes were found to be distributed in two maternal genetic lineages(Mt-Ⅰand Mt-Ⅱ),with the Mt-Ⅰconsisting of three haplogroups(A,B,and E)accounting for 78.95%;the Mt-Ⅱlineage includes two haplogroups(C and D),accounting for 21.05%,suggesting that the Gannan yak has two maternal origins.
