Abstract

In order to analyze the distribution characteristics of skin microbiota diversity in captive Western Pacific spotted seals(Phoca largha),skin swab samples were collected from the seals at different status under captive environment to extract total nucleic acids,and then to detect the abundance and types of microbiota contained therein using high-throughput sequencing.The results showed that 23 phyla and 359 genera were involved by the microbiota,including Proteobacteria(73.12%)and Bacteroidetes(21.28%)with relatively high abundance,Psychrobacter(57.66%)and Ornithobacterium(17.74%)that were dominant;the microbial diversity was similar in lesioned and healthy skin,where the abundance of core microbiota changed.In the lesioned skin,Proteobacteria and Psychrobacter reduced,in contrast,Ornithobacterium increased.In conclusion,the skin microbiota in captive Western Pacific spotted seals(Phoca largha)was highly diverse,with certain commonalities and specificities in those at different status under same captive environment.A theoretical basis was provided by the study for future diagnosis and treatment of skin diseases in captive seals.
