Evaluation on 4 Common Detection Methods for Brucella Antibodies

Abstract
In order to compare the performance of single and vertical testing(primary screening and confirmation)of common detection methods for Brucella antibodies,including Rose Bengal plate agglutination test(RBT),standard-tube agglutination test(SAT),indirect ELISA(iELISA)and competitive ELISA(cELISA),Brucella antibody-positive sera from cattle and sheep,as well as 275 clinical serum samples from these animals were singly and vertically tested by these methods to assess their analytical sensitivity,analytical specificity,positive detection rate and concordance rate.The results showed that the minimum detection limits of RBT,SAT,iELISA and cELISA for bovine sera were 25.0,100.0,3.1 and 3.1 IU/mL,respectively,and for ovine sera were 25.0,50.0,50.0 and 12.5 IU/mL,respectively;the overall positive detection rates for clinical serum samples by the 4 single methods were ranked as cELISA(70.91%)>iELISA(66.55%)>RBT(39.64%)>SAT(36.00%);and iELISA screening+cELISA confirmation(61.09%)>RBT screening+cELISA confirmation(39.64%)>iELISA screening+SAT confirmation(36.00%)>RBT screening+SAT confirmation(34.55%)by the vertical method;for the single methods,the consistency between agglutination methods(RBT and SAT)as well as that between ELISA methods(iELISA and cELISA)were very strong and high,respectively;for vertical methods,the consistency using agglutination methods for screening or confirmation was very strong,and that between above-mentioned combination and iELISA screening+cELISA confirmation was moderate.In conclusion,the sensitivity and concordance of the 4 methods were different when they were used separately or jointly,requiring to select a suitable method as appropriate.关键词
