Abstract

Novel duck reovirus infection(NDR)is an infectious disease with high morbidity and high mortality rate in ducklings caused by NDRV,mainly resulting in necrosis of diseased duck liver and spleen.In order to develop a safe and efficient new vaccine for the prevention of NDRV infection,the prokaryotic expression plasmid pET28a-NDRV-I3-σB/σC of the σB/σC protein of NDRV-CD strain was con-structed,and the recombinant protein was purified,and the subunit vaccine based on the I3-01 vector was prepared in combination with the ISA 201 VG biphasic adjuvant of Sebic,and the 1-day-old ducklings were divided into I3-σB immunization group,I3-σC immunization group,whole virus inactivation group and control group for immune protection evaluation.The results showed that the antibody titer of the I3-σC immunization group was significantly higher than that of the control group after the first immu-nization,and the antibody titer continued to increase after the interval of 13 days,and was signifi-cantly higher than that of the I3-σB immunization group and the whole virus inactivated group.The hep-atosplenic viral load and anal swab excretion volume in the I3-σC immunization group were significant-ly lower than those in the I3-σB immunization group and the whole virus inactivation group 7 days after the challenge.There was no liver and spleen lesions in the I3-σC immune group,and the protective ef-fect was the best.The results indicated that the I3-σC protein vaccine was superior to the I3-σB pro-tein vaccine and the inactivated vaccine by inducing highly effective neutralizing antibodies and in-hibiting viral replication.This trial provides a theoretical basis for the development of NDRV subunit vaccines,and it is necessary to optimize the immunization schedule and explore multivalent vaccine strategies to improve the application potential in the future.
