PEDV N蛋白重组减毒沙门菌SC3(pYA3342-N)的构建及免疫效果评价
Construction of a recombinant attenuated Salmonella strain SC3(pYA3342-N)of PEDV N protein and evaluation of its immune effect

Abstract
To preliminarily evaluate the vaccine potential of the porcine epidemic diarrhea virus nucleocapsid(PEDV N)gene,this study utilized recombinant attenuated Salmonella to express the N protein and assessed immune responses in immunized mice.The N gene from the PEDV LJX strain was cloned into the pYA3342 vector via homologous recombination and electroporated into attenuated Salmonella SC3,generating the recombinant strain SC3(pYA3342-N).Stable genetic expression of PEDV N in SC3 was confirmed by RT-PCR and Western-blot.Following safety validation,7-week-old male BALB/c mice were immunized for immune evaluation.RT-qPCR analysis revealed significantly elevated IFN-γ levels in the SC3(pYA3342-N)and SC3 groups compared to the blank control group,indicating bacterial colonization in intestinal tissues and subsequent induction of cellular immunity.ELISA results demonstrated that SC3(pYA3342-N)-immunized mice exhibited markedly higher serum IgG and mucosal sIgA antibody levels than both the SC3 and control groups,with serum IgG titers showing a dose-dependent increase upon booster immunizations.These findings collectively indicate that the recombinant attenuated Salmonella SC3(pYA3342-N)effectively triggers robust cellular,humoral,and mucosal immune responses.This study provides preliminary evidence supporting PEDV N as a promising vaccine candidate and lays the foundation for developing oral live-vectored vaccines against PEDV.关键词
