Establishment of a new method for increasing Salmonella in eggs and its application in the detection of Sm24/Rif12/Ssq live vaccine for reducing the contamination rate of free-range laying hens

Abstract
This study aims to develop a novel method for enhancing the detection of Salmonella in eggs and to evaluate its application in reducing the contamination rate of free-range laying hens vaccinated with the live vaccine Sm24/Rif12/Ssq.To improve the sensitivity of detection for low-level contamination of Salmonella and to resuscitate bacteria in a viable but non-culturable(VBNC)state,TSB pre-enrichment broth was supplemented with 35 μg/mL ferrous sulfate and 2 mmol/L sodium pyruvate.Over a six-month period(from August 2023 to January 2024),40 eggs were randomly sampled every 15 days(a total of 480 eggs)for Salmonella isolation and identification.Additionally,at each sampling time,10 environmental samples were collected using cotton swabs moistened with 0.1%peptone water from the chicken coop.The results demonstrated that the modified TSB(mTSB)significantly enhanced the detection rate of Salmonella in eggs and exhibited superior recovery of Salmonella in the VBNC state compared to standard TSB enrichment(P<0.01).No Salmonella was detected in the eggs or eggshells of free-range hens vaccinated with the Sm24/Rif12/Ssq vaccine.This study confirms that mTSB is effective for isolating Salmonella with low contamination levels and VBNC status in eggs.Furthermore,the live Salmonella Enteritidis vaccine Sm24/Rif12/Ssq reduces the risk of Salmonella contamination in free-range local breed eggs,providing a reliable strategy for ensuring the safety of egg products in free-range laying hens.关键词
