一株新型鸭呼肠孤病毒的分离鉴定及致病性分析
Isolation,identification and pathogenicity analysis of a novel duck reo virus

Abstract
To investigate the prevalence of novel duck reovirus(NDRV),a viral strain was success-fully isolated using duck embryo propagation methods from the liver and spleen tissues of ducks suspected to be infected with NDRV in Weifang,Shandong Province.The isolate was confirmed as NDRV through RT-PCR,next-generation sequencing,homology comparison,and phylogenetic analysis of the S1 gene segment.The complete genome measured 23 419 bp in length,with the S1 gene encoding the P10,P18,and σ C proteins.Phy-logenetic analysis demonstrated that the isolate clustered within the NDRV reference strains available in the NCBI,sharing the highest nucleotide homology(98.1%—99.8%)with the NDRV J18 strain.The isolate was designated as NDRV-SDWF-01.To evaluate viral replication,the isolate was inoculated into leghorn male hepatoma(LMH)cells and serially passaged to the 5th generation(F5).Viral replication was confirmed by indirect immunofluorescence assay(IFA),with TCID50 determined as 1× 10-6.4/0.1 mL.Growth curves in BHK21,Vero,and LMH cells revealed superior adaptation to LMH cells,evidenced by specific green fluorescence.To further investigate the pathogenicity of the NDRV-SDWF-01 strain,3-day-old cherry valley ducklings were injected with 0.2 mL of F5 virus solution intramuscularly in the legs.The infected ducklings exhibited significant growth retardation,with their body weight consistently lower than that of the negative control group from day 3 post-inoculation onward.By day 7,one duckling succumbed to infection(10%mortality).Viral shedding was detectable as early as day 1 post-inoculation,peaked on day 5,and gradually declined thereafter.The results provide some reference for the analysis of NDRV genetic evolution and disease prevention and control.关键词
